Preparing the Stock Solutions Required for Running Tricine SDS Gels

  • Prepare cathode buffer stock solution.
    • In a large bottle combine tris, tricine, 20% SDS.
    • Bring up to required volume with H2O. The pH works out itself.
  Cathode buffer stock (5X) - 1 L Cathode buffer stock (5X) - 3 L
Tris 60g 180g
Tricine 90g 240g
20% SDS 25 mL (or 5g/L) 15g
H2O to 1L to 3L
  • Prepare anode buffer stock solution.
    • Pour tris Into a large bottle.
    • Bring pH to 8.9 by adding HCl and testing with pH meter.
    • Add H2O to required volume.
  Anode buffer stock (10X) - 1 L
Tris 240g
HCl pH to 8.9 (approx 35-40mL)
H2O to 1 L
